I know that the BATFE requires that the local officials confirm that the proposed location complies with local zoning laws. I've seen people asking the city council for a variance to allow them to obtain a FFL. The zoning administrator has explained that the need for the variance came up when the BATFE called him to find out if the zoning is proper.
I don't know anything about requirements for sepeate entrances, etc. Prior to even considering an FFL, I had a seperate shop built into my detached garage. This shop has safes in it, is climate controlled, and is alarmed with a radio dispatch so that it doesn't require a telephone or cable line to dispatch police. I have always used this area for my FFL business, and it is the area where I showed the initial inspector when I applied. He never asked any more questions that I can recall, but that was several years ago.
You have to have an FFL to do gunsmith work too, and most gunsmiths work out of their residence it seems. It is not impossible to get a home based FFL, but it can be difficult and will likely become more difficult. I got mine under Slick Willie, so I'm somewhat used to the tactics to put screws to the little guy, but with Barry at the helm and someone like Chuck Schumer as AG and a like minded head of the BATFE, things could get downright scary.

But in my dealings with the ATF if you called them now and asked to set up an FFL shop at your home. HMMM. Good luck.
